課題組簡介

本課題組由張俊武研究員建立于1993年,承擔(dān)過國家七.五、863、基礎(chǔ)研究重大前期專項、973、重大科學(xué)計劃、美國中華醫(yī)學(xué)基金(CMB)、國家自然科學(xué)基金重大項目和面上項目等,在1990-2010近二十年間重點研究珠蛋白基因表達調(diào)控、造血相關(guān)基因功能及機制及老年性癡呆(AD)相關(guān)基因,已畢業(yè)博士33人,碩士6人。共發(fā)表論文130多篇(SCI 44篇)。2012年,余佳研究員接任課題組長,聚焦“RNA和干細(xì)胞生物學(xué),重點關(guān)注RNA和遺傳信息傳遞如何影響干細(xì)胞命運決定、造血譜系分化及相關(guān)疾病,以期為人類疾病的細(xì)胞治療、靶向治療及成分輸血提供理論基礎(chǔ)和新方向。通過近十年努力,課題組建立了成熟的干細(xì)胞培養(yǎng)及移植平臺、RNA實驗平臺、組學(xué)及計算平臺;培養(yǎng)了一批科研骨干及生物信息學(xué)團隊;并與國內(nèi)外多個領(lǐng)域內(nèi)一流實驗室建立了緊密合作關(guān)系、共享研究資源、共同培養(yǎng)研究生與博士后。現(xiàn)有工作人員包括:余佳(教授)、 張俊武(研究員)、王芳(教授)、馬艷妮(研究員)、王小爽(副研究員)、趙華路(副主任技師)、徐嘉悅(助理研究員)、高玉風(fēng)(助理研究員)、司艷敏(技術(shù)員)、果月紅(技術(shù)員)、博士后4人(劉思琪、李衛(wèi)倩、何家驩、孫晨光),在讀博士研究生11人,碩士研究生10人。課題組既有豐富的歷史沉淀,又年輕而朝氣蓬勃,學(xué)術(shù)氣氛積極向上,致力于成為嚴(yán)謹(jǐn)、求實、快樂、奮進的研究團隊。歡迎各位有志于科學(xué)研究、對干細(xì)胞及RNA生物學(xué)感興趣的考生報考。

個人簡介

余佳,男,1978年生,研究員,博士生導(dǎo)師,北京協(xié)和醫(yī)學(xué)院長聘教授(首批)、RNA與干細(xì)胞生物學(xué)課題組組長,中國醫(yī)學(xué)科學(xué)院RNA與造血調(diào)控研究重點實驗室主任,醫(yī)學(xué)分子生物學(xué)國家重點實驗室PI。獲得國家杰出青年科學(xué)基金(2017,生命科學(xué)部),長江學(xué)者獎勵計劃(2017),百千萬人才工程國家級人選(2019),國務(wù)院政府特殊津貼(2020)等人才項目支持。擔(dān)任科技部十三五發(fā)育編程及其代謝調(diào)節(jié)重點研發(fā)專項首席科學(xué)家,十四五發(fā)育編程及其代謝調(diào)節(jié)重點研發(fā)專項總體專家組、指南編寫組成員。20067月畢業(yè)于中國醫(yī)學(xué)科學(xué)院/北京協(xié)和醫(yī)學(xué)院基礎(chǔ)醫(yī)學(xué)研究所生化與分子生物學(xué)系,理學(xué)博士。2006-2007年在美國伊利諾伊大學(xué)芝加哥分校工作(Post-doc)2007-2009年在美國西北大學(xué)Feinberg醫(yī)學(xué)院工作(Post-doc)200910月回國加入中國醫(yī)學(xué)科學(xué)院基礎(chǔ)醫(yī)學(xué)研究所生化系及醫(yī)學(xué)分子生物學(xué)國家重點實驗室。作為負(fù)責(zé)人承擔(dān)國家自然科學(xué)基金委重大研究計劃、重點項目及面上項目等;作為負(fù)責(zé)人承擔(dān)科技部國家重點研發(fā)計劃、973子課題及國家重大研究計劃。余佳博士同時擔(dān)任中國醫(yī)學(xué)科學(xué)院-以色列Weizmann研究所聯(lián)合實驗室PI,中國醫(yī)學(xué)科學(xué)院醫(yī)學(xué)與健康科技創(chuàng)新工程創(chuàng)新團隊項目首席科學(xué)家,人血細(xì)胞分子圖譜(ABC)研究聯(lián)盟首批倡議及參加成員。余佳實驗室主要興趣集中在造血調(diào)控、干細(xì)胞命運及轉(zhuǎn)化醫(yī)學(xué),綜合運用多種組學(xué)及計算生物學(xué)方法,干細(xì)胞培養(yǎng)及基因操縱等體外實驗技術(shù),干細(xì)胞移植、基因敲除小鼠及譜系示蹤等體內(nèi)技術(shù),結(jié)合生化與分子生物學(xué)手段,研究RNA、互作蛋白質(zhì)及相關(guān)遺傳信息傳遞過程在干細(xì)胞或疾病中的功能和調(diào)控機理。在組織器官發(fā)育與疾病的細(xì)胞圖譜、非編碼RNA生成與加工新機理、干細(xì)胞命運調(diào)控新機制等方面取得突出成果,相關(guān)研究成果作為通訊作者(含共同)在Cell Stem CellDevelopmental CellBloodCell ResearchScience AdvancesEMBO Journal20152021)、Genome ResearchNature Communications20172019)、Genome Biology Cancer Research201020152020)、Nucleic Acids Research2013201420172020)、AutophagyOncogeneMCB等國際主流學(xué)術(shù)期刊上發(fā)表SCI論文30余篇,被引用4000余次。獲教育部自然科學(xué)二等獎(2018)。現(xiàn)擔(dān)任中國生理學(xué)會血液生理專業(yè)委員會理事,中國細(xì)胞生物學(xué)會干細(xì)胞專業(yè)委員會理事, 北京市生化協(xié)會理事。擔(dān)任Blood, Nature Communications, Nucleic Acids Research, Oncogene, Cancer Research, MCB, JBC等雜志特邀審稿人。

研究工作簡介

              RNA生物學(xué)是系統(tǒng)認(rèn)識和研究各種RNA的生物發(fā)生、分子特征、生物學(xué)功能和分子機理的一門學(xué)科,也是生命科學(xué)領(lǐng)域的新興學(xué)科。過去二十年里,這一學(xué)科得到長足發(fā)展,特別是高通量測序技術(shù)的出現(xiàn)不但幫助發(fā)現(xiàn)了更多編碼RNA的特征,而且鑒定了多種非編碼RNA分子,是目前國際上最前沿的領(lǐng)域之一。干細(xì)胞生物學(xué)也是目前國際上最前沿和挑戰(zhàn)最大的領(lǐng)域之一。闡明干細(xì)胞自我更新和分化的調(diào)控機理可為再生醫(yī)學(xué)和癌癥治療提供重要支撐。以RNA為切入點研究干細(xì)胞生物學(xué)具有獨特優(yōu)勢:i. 以往被視為遺傳信息傳遞中間站的messenger RNA (mRNA)其實具有遠超我們所理解的轉(zhuǎn)錄后調(diào)控方式,如RNA編輯、RNA修飾、RNA穩(wěn)定性調(diào)節(jié)、RNA選擇性剪接及RNA選擇性加帽或加尾等,這些方式均會引起相應(yīng)的基因表達變化,從而最終改變遺傳信息從DNA向蛋白質(zhì)的共線性傳遞。近期關(guān)于RNA修飾的研究已經(jīng)衍生出RNA表觀遺傳學(xué)這一學(xué)科方向,也誕生了表觀轉(zhuǎn)錄組(Epitranscriptome)概念;ii. RNA組學(xué),特別是單細(xì)胞RNA組學(xué)的發(fā)展,使我們可以精確描繪發(fā)育層級的分子動力學(xué)圖譜,從而推動了目前中國、美國及歐洲的人類細(xì)胞圖譜計劃;iii. ENCODE計劃最重要的貢獻之一:非編碼RNA的大量發(fā)現(xiàn),更是大大擴充了人類對于自身,乃至各個物種基本遺傳和進化的認(rèn)知。但該計劃也提出了未來多個重要的挑戰(zhàn),即各種各樣的非編碼RNA的分子特征是什么,是否具有重要的生物學(xué)功能,有什么獨特的分子調(diào)控機制,在轉(zhuǎn)化醫(yī)學(xué)中是否有應(yīng)用價值?

課題組主攻下列科學(xué)問題:

1、造血干細(xì)胞起源、異質(zhì)性、譜系分化與生態(tài)的分子動力學(xué)圖譜;

2、非編碼RNA加工與成熟的分子機制及其在干細(xì)胞分化與相關(guān)疾病中的作用機理;

3、胚胎及成體干細(xì)胞功能的RNA調(diào)控機理;

4RNA代謝及調(diào)控的生理功能和轉(zhuǎn)化醫(yī)學(xué)應(yīng)用。

馬艷妮,女,198111月出生,北京協(xié)和醫(yī)學(xué)院準(zhǔn)聘助理教授,研究員,士生導(dǎo)師。20087月畢業(yè)于中國醫(yī)學(xué)科學(xué)院基礎(chǔ)醫(yī)學(xué)研究所生化與分子生物學(xué)系,理學(xué)博士。2008-2011年于中國科學(xué)院生物物理研究所任助理研究員。20117月加入中國醫(yī)學(xué)科學(xué)院基礎(chǔ)醫(yī)學(xué)研究所生化與分子生物學(xué)系,醫(yī)學(xué)分子生物學(xué)國家重點實驗室工作。作為負(fù)責(zé)人承擔(dān)國家自然科學(xué)基金委面上、青年項目,作為科研骨干參與科技部973項目、國家重點研發(fā)計劃。獲國家優(yōu)秀青年科學(xué)基金2022)、北京市科技新星”(2017)、醫(yī)科院青年醫(yī)學(xué)人才獎勵項目2018)、 協(xié)和新星” (2015)等人才計劃支持。作為第一作者或通訊作者(含共同)在Cell Stem CellDevelopmental CellBloodEMBO J20152021)、Genome ResearchNature CommunicationsMol Cell Bio等國際學(xué)術(shù)期刊發(fā)表SCI論文14篇。

個人簡介

高玉風(fēng),女,198911月生,博士,助理研究員。20186月畢業(yè)于中國科學(xué)院北京基因組研究所,基因組學(xué)專業(yè),理學(xué)博士。20187月加入中國醫(yī)學(xué)科學(xué)院基礎(chǔ)醫(yī)學(xué)研究所生化與分子生物學(xué)系,醫(yī)學(xué)分子生物學(xué)國家重點實驗室工作。作為負(fù)責(zé)人承擔(dān)國家自然科學(xué)基金委青年基金項目。作為共同第一作者在國際學(xué)術(shù)期刊Blood上發(fā)表研究論文1篇,作為共作者發(fā)表SCI論文2篇。

目前研究方向為利用組學(xué)數(shù)據(jù)和生物信息學(xué)手段描述和刻畫造血干細(xì)胞發(fā)育過程中的分子動態(tài)圖譜,以及疾病中的基因組、轉(zhuǎn)錄組及翻譯組學(xué)研究。

王芳,女,19832月生,教授,博士生導(dǎo)師。2009年畢業(yè)于北京協(xié)和醫(yī)學(xué)院,獲博士學(xué)位,后留所工作至今。2012年晉升副教授,2017年晉升教授,同年獲聘博士生導(dǎo)師。作為負(fù)責(zé)人承擔(dān)國家自然科學(xué)基金委青年、面上項目,作為科研骨干參與科技部973項目,國家重點研發(fā)計劃。獲科技部重點研發(fā)計劃青年科學(xué)家項目 (2022)國家優(yōu)秀青年科學(xué)基金2021)、醫(yī)科院青年醫(yī)學(xué)人才獎勵項目2018)、 協(xié)和新星” (2016)等人才計劃支持。作為第一作者或通訊作者(含共同)在Nature Cell BiologyCell ResearchNature CommunicationsGenome BiologyCancer ResearchNucleic Acids ResearchAutophagyOncogeneJBC等國際學(xué)術(shù)期刊發(fā)表SCI論文20篇。獲教育部自然科學(xué)二等獎(2018)。
